The role of the ubiquitin-proteasome system in tumorigenesis and cancer treatment.
Regulated protein degradation is mediated by ubiquitin-proteasome system. Inhibition of proteasome proteolytic activity is a promising approach in cancer therapy with first-in-class drug bortezomib approved for the treatment of patients with multiple myeloma. Since ubiquitination regulates not only protein degradation but also wide variety of cellular signaling pathways, this new approach might bring new drugs with better efficacy and safety profiles.
