Implications of epigenetic changes in cells derived from prostate cancer
The thesis is focused on a study of DNA methyltransferase (5-aza-2´-deoxycytidine, Aza-dC) and histone deacetylase (sodium butyrate, NaB) inhibitors effects on prostate cancer cells with modified expression of the androgen receptor gene. The androgen-independent human prostate cancer cell lines PC3 (without a functional androgen receptor) and DU145 (with strongly limited expression due to methylation of the androgen receptor gene) are used. The methodology is based on PCR of bisulfite-modified DNA and RT-PCR with bisulfite-sequencing.
